Evaluation of a Series of Different Surface Charged Fullerenol Nanoparticles as Reactive Oxygen Species Scavengers and Potential Cytoprotective Agents

摘要Purified fullerenol nanoparticles (NPs) with isoelectric points (pIs) 7.0, 5.29, 4.40 and 2.81 were used to scavenge 2,2-diphenyl-1-picryhydrazyl (DPPH) in vitro and determine the effect on cytotoxicity in Raw 264.7 cells. The cell viabilities treated with fullerenol NPs among the pI 7.0 to 2.81 were analyzed by statistical product and service solutions. The results showed that their cytoprotective abilities and scavenging radical DPPH were in the order of pI 7.0 > pI 5.29 > pI 4.40 > pI 2.81, respectively. Fullerenol NPs with pI 2.81 was less cytoprotective, which could be caused by the weakest radical-scavenging capabilities among the four samples. It could be due to 1,3-tautomeric shift of the hydroxyl hydrogen to produce the most electronegative C O groups and to exist in the least C C bonds on fullerenol molecules with pI 2.81. The differences in free radical-scavenging ability of fullerenol NPs with different pIs support the fact that surface properties (e. g., charge) had influence on the biological activities (i. e., cytoprotective) of fullerenol NPs. The fullerenol NPs with pIs > 2.81 were biocompatible and could be potential cytoprotective reagents.
